Cell Fusion can be performed not only by PEG but also by an Electro Cell Fusion device. Not many researchers are aware that Cell Fusion can be performed by a device! Our LF101 and LF201 devices have been designed for this purpose.
PEG v LF201/LF101 Device
While the majority of researchers employ PEG, there are nevertheless problems associated with such an approach. They include:
- a lack of control on the fusion
- reproducibility being dependant on the skills of the individual researcher
- additionally, PEG presents problems with toxicity
- it can be done by a device
The particular advantage of the LF201/LF101 Device is that:
- in the case of electrofusion and the level of control afforded by the LF201/LF101, the PEG problems do not arise.
- indeed, the LF201/LF101 offers a broad range of capabilities in terms of Electro Fusion for Hybridoma Production
LF201 v LF101
The LF201 has a higher 3ification than the LF101 – it is the next generation model in the series. So, not only does the LF201 perform all the applications that can be done by LF101; it is a high-though-put Electro Cell Fusion Generator. Its wider voltage range enables it to handle a greater number of cells (in its larger chamber) at the same time. In addition, new functions have been added: resistance measurement, bi-polar pulse and attenuating post-fusion AC current etc..

Use of a low voltage is an imperative requirement if one is to obtain successful nuclear transplant. Unlike the LF101 and LF201 many competing devices cannot accurately control voltages in the lower ranges (10V increment setting).
LF201 v LF101
For successful and efficient cell fusion, researchers need to be able to simultaneously handle as large a volume of cells as possible. This meant increasing the size of the cell-fusion chamber from a 2mm gap (in the LF101) to a 3mm gap (in the LF201). Of necessity, a larger gap requires a higher voltage. Hence, why the LF201’s DC voltage range is 1200V and that of the LF101 is 930V. The difference between the two is that the LF201 is designed for the high-through-put cell fusion.

The movie clip (download below) demonstrates an electrofusion device for controlling the precise moment of fusion between droplets by applying an electric field. This device allows:
1. accurate determination of the start of chemical/biological reactions
2. minimum contact of reactants with channel walls – eliminating surface absorption problems
3. easy fabrication and
4. continuous observation of initiated reaction
The movie clip demonstrates the fusion of ß-galactosidase and fluorescein di-ß-D galactopyranoside (FDG) droplets, and observed the enzymatic reaction using fluorescence microscopy. In addition, sequential fusion of pico-litre droplets was also accomplished.
